Vonix is indicated for the following conditions:
- Gastric ulcer
- Duodenal ulcer
- Reflux esophagitis
- Prevention of recurrence of gastric or duodenal ulcer during low-dose aspirin administration
- Prevention of recurrence of gastric or duodenal ulcer during NSAID administration
- Adjunct to Helicobacter pylori eradication in various conditions including gastric ulcer, duodenal ulcer, gastric mucosa-associated lymphatic tissue (MALT) lymphoma, idiopathic thrombocytopenic purpura, the stomach after endoscopic resection of early-stage gastric cancer or Helicobacter pylori gastritis.
Pharmacology: Vonoprazan is a potassium-competitive acid blocker (P-CAB) that inhibits H+, K+-ATPase in a reversible and potassium-competitive manner. It does not require activation by acid and has a strong inhibitory effect on gastric acid production, thereby preventing mucosal damage in the upper gastrointestinal tract.
Dosage:
- Gastric ulcer and duodenal ulcer: 20 mg orally once daily for 8 weeks for gastric ulcer and 6 weeks for duodenal ulcer.
- Reflux esophagitis: 20 mg orally once daily for 4 weeks, extendable up to 8 weeks if needed. Maintenance therapy: 10 mg once daily, increased to 20 mg if efficacy is inadequate.
- Prevention of recurrence of gastric or duodenal ulcer during low-dose aspirin or NSAID administration: 10 mg once daily.
- Adjunct to Helicobacter pylori eradication: 20 mg twice daily along with amoxicillin and clarithromycin for 7 days.
Administration: Vonoprazan can be taken orally without regard to food or the timing of food.
Contraindications:
- Hypersensitivity to Vonoprazan or any excipient
- Concomitant use with atazanavir sulphate, nelfinavir, or rilpivirine hydrochloride.
Side Effects: Common side effects include diarrhea, constipation, drug hypersensitivity, hepatotoxicity, rash, nausea, abdominal distension, and increased liver enzymes.
Pregnancy & Lactation: Use in pregnant or breastfeeding women only if the expected therapeutic benefit outweighs potential risks.
Precautions & Warnings: Close observation of disease progression and minimum necessary dosage is recommended. Caution in the elderly, children, patients with renal or hepatic impairment.
Overdose Effects: No experience of overdose reported. Symptomatic and supportive treatment if overdose occurs.
