Synthox-50 is prescribed for the following conditions:
- Hypothyroidism of any cause, except transient hypothyroidism during the recovery phase of subacute thyroiditis.
- TSH suppression therapy in cases of goiters, nodules, or after thyroid cancer treatment (surgical or radiological).
- To counteract the goitrogenic effects of drugs like lithium.
- As a diagnostic tool in thyroid suppression tests.

Synthox-50 contains synthetic Levothyroxine (T4), which is identical to the natural thyroid hormone produced in the thyroid gland. Key features include:
- Approximately 30% of T4 converts to the active hormone Triiodothyronine (T3) in peripheral tissues.
- T4 binds predominantly to thyroxine-binding globulin (TBG), which prolongs its half-life by protecting it from metabolism and excretion. Only 0.03% of plasma T4 is unbound.
- The half-life of T4:
- Normal conditions: 6–7 days.
- Hyperthyroidism: 3–4 days.
- Hypothyroidism: 9–10 days.
- Conditions like nephrosis, hepatic cirrhosis, or certain drug interactions may shorten T4’s half-life.
- T4 undergoes degradation primarily in the liver and is conjugated with glucuronic acid or sulfate, excreted in the urine, and undergoes enterohepatic recirculation.
- Due to its long half-life, a single daily dose of Synthox-50 ensures stable T3 levels once the correct dosage is established.
