Description

Sinapol 120 mg/5 ml Suspension is a medication that performs its action by obstructing the release of pain and fever chemical messengers.

Paracetamol acts primarily in the central nervous system (CNS) to relieve pain and reduce fever. It increases the pain threshold by inhibiting prostaglandin synthesis and acts on the hypothalamic heat-regulating center to produce antipyresis.

Unlike NSAIDs, it has minimal peripheral anti-inflammatory action. Its exact mechanism involves inhibition of COX enzymes (including COX-1, COX-2, and possibly COX-3), which reduces prostaglandin production. This results in analgesic and antipyretic effects without significant gastrointestinal irritation or platelet inhibition.

Dosage & Administration

Dosage varies by formulation and age:

  • Adults: 1–2 tablets every 4–6 hours; maximum 4 g/day (8 tablets)
  • Children (6–12 years): ½–1 tablet 3–4 times daily
  • Syrup (Children): Dose varies by age and weight, 3–4 times daily
  • Suppository: Used in children and adults based on age and weight
  • IV form: Dose adjusted by body weight and clinical condition

Do not exceed the recommended daily dose.

Drug Interactions

  • Alcohol and enzyme-inducing drugs (barbiturates, anticonvulsants, tricyclic antidepressants) may increase liver toxicity risk or reduce effectiveness.
  • Chronic use of certain medications (e.g., anticonvulsants, oral contraceptives) may increase metabolism of paracetamol, lowering its therapeutic effect.

Contraindications

Sinapol should not be used in patients with:

  • Known hypersensitivity to paracetamol

Side Effects

Generally well tolerated. Rare but possible adverse effects include:

  • Skin rash and allergic reactions
  • Blood disorders (thrombocytopenia, leukopenia, neutropenia, agranulocytosis)
  • Pancreatitis (rare)

Use in Pregnancy & Lactation

Overdose Information

Overdose may cause severe liver injury, which can be life-threatening. Early symptoms include nausea, vomiting, abdominal pain, and loss of appetite. Severe cases may progress to liver failure, kidney damage, metabolic acidosis, or death.

Immediate medical treatment is required. Activated charcoal may be used if taken recently, and N-acetylcysteine is the specific antidote.
