Dasacent (Dasatinib)
- Treatment of adult patients with:
- Newly diagnosed Philadelphia chromosome-positive (Ph+) chronic myeloid leukemia (CML) in chronic phase.
- Chronic, accelerated, or myeloid/lymphoid blast phase Ph+ CML with resistance or intolerance to previous therapy, including Imatinib.
- Philadelphia chromosome-positive acute lymphoblastic leukemia (Ph+ ALL) with resistance or intolerance to previous therapy.
- Treatment of pediatric patients (1 year and older) with Ph+ CML in the chronic phase and newly diagnosed Ph+ ALL in combination with chemotherapy.
Pharmacology:
- Dasatinib inhibits several kinases, including BCR-ABL, SRC family, c-KIT, EPHA2, and PDGFRβ. It helps overcome resistance to Imatinib in leukemic cells by targeting mutations and alternative signaling pathways.
- Absorption: Dasatinib reaches maximum plasma concentrations within 0.5–6 hours after oral administration.
- Distribution: Highly protein-bound (96%) with an apparent volume of distribution of 2505 L.
- Elimination: Half-life is 3-5 hours, and 85% of the dose is excreted in the feces.
- Metabolism: Primarily metabolized by CYP3A4, with the active metabolite accounting for 5% of exposure.
