Indications

A prescription drug called Don A 10 is used to treat nausea, vomiting, and indigestion. It enhances food movement in the stomach, which helps with digestion, and blocks the brain chemical that causes nausea or vomiting. Don A 10 is taken in the prescribed dose and time frame before meals.

Pharmacology

Don-A is a dopamine antagonist that principally blocks the dopamine receptors located in the ChemoreceptorTrigger Zone (CTZ) and stomach.

Dosage & Administration

Don-A should be taken 15-30 minutes before meals and, if necessary, before retiring.

Interaction

may counteract bromocriptine’s hyperprolactinaemic effects. antimuscarinics and opioid analgesics may counteract the prokinetic effect. Possible Fatal: Strong CYP3A4 inhibitors, such as ritonavir, erythromycin, or ketoconazole, may raise serum domperidone levels, raising the risk of QT prolongation in the process.

Contraindications

Don-A is contraindicated to patients having known hypersensitivity to this drug and in the case of neonates.

Side Effects

Don-A may produce hyperprolactinemia (1.3%). This may result in 

• Drowsiness
• Extrapyramidal reactions
• Decreased libido
• Skin rash,
• Itch.
• Lassitude
• Galactorrhea,
• Breast enlargement, Soreness, and
• Reduced libido.

Pregnancy & Lactation

The safety of Don-A has not been proven and it is therefore not recommended during pregnancy.

Precautions & Warnings

• Don-A is highly metabolized in the liver, it should be used with caution in patients with hepatic impairment.
• Inform your doctor if you get watery diarrhea, fever, or stomach pain that does not go away.
• Do not take it for longer than 7 days without consulting your doctor.
