Loperamide-Simethicone Tablets, This medication is indicated for the symptomatic treatment of acute diarrhea in adults and adolescents (12 years and older) when associated with gas-related discomfort such as bloating, cramping, or flatulence.
- Loperamide binds to opioid receptors in the gut wall, slowing peristalsis, increasing intestinal transit time, and enhancing water and electrolyte absorption. It also improves anal sphincter tone to reduce urgency.
- Simethicone acts as an anti-foaming agent, breaking down gas bubbles in the gastrointestinal tract, which helps relieve bloating and discomfort.
- Loperamide undergoes extensive first-pass metabolism, with a bioavailability of approximately 0.3% and 95% plasma protein binding. It is primarily metabolized in the liver and excreted via bile and feces, with a half-life of about 11 hours.
- Simethicone is not absorbed and works locally in the gut.
