Indications

Lacidipine is used to treat hypertension alone or in conjunction with other antihypertensive medications such as beta-blockers, diuretics, and ACE inhibitors.

 

Pharmacology

Lacidipine works by competitively inhibiting voltage-dependent calcium ‘L’ channels, which prevents calcium from entering smooth muscle cells. The actions produced by the influx of calcium into the cells, such as the activation of contractile proteins in smooth muscle, are inhibited as a result of this activity.

 

Dosage and Administration

Lacidipine is best taken once a day at a dose of 2 mg. After 3 to 4 weeks, the dose can be increased to 4 mg and then, if necessary, to 6 mg. Lacidipine is best taken first thing in the morning.

 

Interaction

Lacidipine plasma concentrations can be boosted by taking cimetidine at the same time. In investigations using common antihypertensive drugs, as well as digoxin, tolbutamide, and warfarin, no significant pharmacodynamic interaction issues were discovered.

 

Contraindications

Cardiogenic shock, unstable angina, aortic stenosis, avoidance within 1 month of myocardial infarction, reduced left ventricular function are all reasons to avoid this product.

 

Side Effects

Lacidipine is well tolerated by most people. Minor side effects such as flushing, palpitation, edema, headache, dizziness, and infrequently gastro-intestinal disturbances, gum hyperplasia, mood disorders, asthenia, polyuria, muscle cramps, skin rash, and others may occur.

 

Pregnancy & Lactation

Lacidipine and pregnancy: There is no information on the safety of Lacidipine during pregnancy in humans. Lacidipine has not been shown to have a teratogenic impact in animal studies.

Lactation: Lacidipine (or its metabolites) is likely to be excreted in breast milk, according to animal milk transfer studies. Lacidipine, on the other hand, has no clinical expertise in pregnancy or lactation. Lacidipine should therefore not be taken during pregnancy or lactation.

Precautions & Warnings

Abnormal cardiac conduction, low cardiac reserves, hepatic impairment; discontinue if ischemia discomfort begins soon after starting medication or if cardiogenic shock develops; avoid grapefruit juice.
