Magolin Tablet (Mirogabalin) is prescribed for the management of neuropathic (nerve-related) pain, including:
- Diabetic Peripheral Neuropathic Pain (DPNP)
- Postherpetic Neuralgia (PHN)
- Peripheral Neuropathic Pain (PNP)
Mirogabalin is a gamma-amino acid derivative that selectively binds to the α2δ subunits of voltage-gated calcium channels (VGCCs). By reducing calcium influx into nerve cells, it decreases the release of pain-signaling neurotransmitters, helping to reduce nerve pain and calm overactive nerve activity.
After oral administration, Mirogabalin is rapidly absorbed, with peak blood levels reached within 0.5–1.5 hours. It has low plasma protein binding (approximately 25%) and is primarily eliminated unchanged through the kidneys. A small portion is metabolized by the liver. Its average elimination half-life ranges from 2 to 5 hours.
Dosage & Administration
Adults
- Initial dose: 5 mg twice daily
- Increase by 5 mg per dose at intervals of at least one week
- Usual maintenance dose: 10–15 mg twice daily, depending on the patient’s condition and response.
Children & Adolescents
- Safety and effectiveness have not been established.
Hepatic Impairment
- No significant adverse effects have been observed after a single 15 mg dose in patients with mild to moderate liver impairment.

Renal Impairment
- Mild impairment: Start with 5 mg twice daily, increase gradually to 10 mg twice daily.

Drug Interactions
With medicines
- Cimetidine and Probenecid may increase Mirogabalin blood levels.
- Lorazepam may enhance the sedative effects of Mirogabalin.
- Medicines that inhibit OAT1, OAT3, OCT2, MATE1, MATE2-K, or UGT transporters may increase Mirogabalin exposure and should be used cautiously.
With food and alcohol
- Food does not significantly affect absorption.
- Avoid alcohol, as it can increase drowsiness, dizziness, and impair coordination.
Contraindications
Do not use Magolin if you have:
- Hypersensitivity to Mirogabalin or any of its ingredients.
- Mild to moderate hepatic or renal impairment (as stated in the provided source).
