Pharmacology
Pregabalin is a structural derivative of gamma-aminobutyric acid, an inhibitory neurotransmitter (GABA). It does not directly bind to the GABAA, GABAB, or benzodiazepine receptors. Pregabalin has a high affinity for the alpha-2-delta site in the central nervous system, which is a voltage-gated calcium channel auxiliary subunit. Pregabalin has a dose-independent oral bioavailability of 90%. Its half-life is 6.3 hours in individuals with normal renal function, and the kidneys are primarily responsible for excreting it from the systemic circulation as an unaltered medication.
Dosage & Administration of Pregaba 75
Neuropathic pain associated with diabetic peripheral neuropathy:Â
- The maximum dose of Pregabalin that is advised for patients with creatinine clearance of at least 60 mL/min is 100 mg three times per day (300 mg/day). The dosage should start at 50 mg three times per day (150 mg/day), and depending on effectiveness and tolerability, it may be increased to 300 mg/day within a week.
Post-herpetic neuralgia:Â
- For patients with a creatinine clearance of at least 60 mL/min, Pregabalin has dosed at 75 to 150 mg twice daily or 50 to 100 mg three times daily (150 to 300 mg/day). The dosage should start at 75 mg twice daily or 50 mg three times daily (150 mg/day), and depending on effectiveness and tolerability, it may be increased to 300 mg/day within a week.
Adjunctive therapy for adult patients with partial onset seizures:Â
- In general, it is advised that patients start treatment with a daily dose cap of no more than 150 mg (75 mg two times a day, or 50 mg three times a day). The dose may be increased to a maximum of 600 mg/day depending on how each patient responds and tolerates it.
Management of Fibromyalgia:Â
- The recommended dose of Pregabalin for fibromyalgia is 300 to 450 mg/day. Dosing should begin at 75 mg two times a day (150 mg/day) and may be increased to 150 mg two times a day (300 mg/day) within 1 week based on efficacy and tolerability.
Neuropathic pain associated with spinal cord injury:Â
- The suggested dosage ranges from 150 to 600 mg per day. The starting dosage is 75 mg twice daily, or 150 mg per day. Depending on effectiveness and tolerability, the dose may be increased within a week to 150 mg twice daily (300 mg/day).
Pregabalin capsules can be taken without regard to meals.
Interaction of Pregaba 75
Pregabalin has no noteworthy interactions with other antiepileptic medicines or oral contraceptives. The effects of ethanol and lorazepam may be amplified by pregabalin.
Contraindications
Patients who have a known hypersensitivity to Pregabalin or any of its components should avoid taking it.
Side Effects of Pregaba 75
Dizziness, somnolence, dry mouth, edema, blurred vision, weight gain, and aberrant thinking are the most common adverse effects.
Pregnancy & Lactation
Pregabalin has a pregnancy category of C. As a result, it should only be taken if the potential benefit outweighs the dangers to the fetus. Pregabalin, like other medicines, can pass into breast milk, thus it should only be used in nursing mothers if the benefits clearly outweigh the risks.
Precautions & Warnings of Pregaba 75
- Without tapering, pregabalin withdrawal may result in insomnia, nausea, headaches, and diarrhea.Â
- As a result, it should be tapered gradually over a minimum of one week rather than stopping suddenly.Â
- Your creatinine kinase level could increase if you take Pregabalin.Â
- It should be stopped right away if myopathy is found or suspected, or if creatinine kinase levels are unusually high.
