Tofacit 5 is prescribed for adult patients with moderately to severely active rheumatoid arthritis (RA) who have shown an insufficient response or intolerance to methotrexate. It may be used:
Alone (monotherapy) Or in combination with methotrexate or other non-biologic DMARDs (disease-modifying antirheumatic drugs).
- Not recommended during pregnancy or breastfeeding.
- Inform your doctor if you are pregnant, planning pregnancy, or nursing.
- Tofacit 5 is a Janus kinase (JAK) inhibitor.
- JAKs are enzymes that transmit signals from cytokines and growth factors to promote inflammation and immune cell activity.
- Tofacitinib disrupts the JAK-STAT signaling pathway, preventing the activation of STATs, which are responsible for gene expression related to inflammation.
- By blocking this pathway, it reduces proinflammatory cytokine production, thus alleviating inflammation and slowing joint damage in RA.
Tofacit 5 may lower the body’s ability to fight infections.
- Report any signs of infection (e.g., fever, cough, chills, muscle aches) to your healthcare provider promptly.
- Avoid live vaccines during treatment.
- Caution in patients with:
- A history of recurrent infections
- Liver or kidney impairment
- Increased risk of thrombosis
