Xerova 10 mg is indicated as an adjunct to diet and lifestyle modification when non-pharmacological measures are insufficient, for the following conditions:

  • Primary hypercholesterolaemia
  • Heterozygous and homozygous familial hypercholesterolaemia
  • Mixed dyslipidaemia (Fredrickson Types IIa and IIb)
  • Hypertriglyceridaemia (Fredrickson Type IV)
  • Dysbetalipoproteinaemia (Fredrickson Type III)

Reduction of cardiac ischaemic events in patients with asymptomatic or mild-to-moderate coronary artery disease and elevated LDL-cholesterol

Hypercholesterolaemia associated with diabetes mellitus or renal transplantation

Atorvastatin is a liver-selective, competitive inhibitor of HMG-CoA reductase, the rate-limiting enzyme involved in the conversion of HMG-CoA to mevalonate, a critical precursor in cholesterol biosynthesis. By suppressing hepatic cholesterol synthesis, atorvastatin effectively lowers elevated LDL-cholesterol levels, leading to a significant reduction in coronary events and mortality associated with coronary heart disease.

Atorvastatin decreases plasma cholesterol and lipoprotein concentrations by inhibiting HMG-CoA reductase and enhancing hepatic LDL receptor expression, thereby increasing LDL uptake and catabolism. It also reduces LDL particle production and count. In some patients, statin therapy modestly increases HDL-cholesterol, further contributing to cardiovascular risk reduction.

Although HMG-CoA reductase does not directly regulate triglyceride metabolism, atorvastatin significantly reduces triglyceride levels in patients with hypertriglyceridemia. This effect is attributed to reduced VLDL synthesis and secretion due to decreased cholesterol availability, as well as enhanced clearance of VLDL remnants via upregulated LDL receptors.
