Indications

The antibiotic nitazoxanide is used to treat Cryptosporidium parvum, Giardia lamblia, and Entamoeba histolytica diarrhea.

Pharmacology

Nitazoxanide is a synthetic antiprotozoal drug that is taken orally. The antiprotozoal activity of Nitazoxanide is thought to be due to interference with the enzyme-dependent electron transfer process Pyruvate Ferredoxin Oxido Reductase (PFOR). This process is required for protozoa’s anaerobic energy metabolism. In vitro, nitazoxanide and its metabolites, diloxanide, suppress the growth of Cryptosporidium parvum sporozoites, oocysts, and Giardia lamblia trophozoites.

Dosage & Administration

Age 1-3 years: 1 tea spoonful or 5 ml suspension every 12 hours for 3 days.

Age 4-11 years: 2 tea-spoonfuls or 10 ml suspension every 12 hours for 3 days.

Age 12 years or above: 5 tea-spoonfuls (25 ml) suspension or 1 tablet every 12 hours for 3 days.

It is recommended to be administered with food.

 

Interaction

It is highly bound to plasma protein. Therefore, caution should be exercised when administering Nitazoxanide concurrently with other highly plasma protein-bound drugs with narrow therapeutic index.

 

Contraindications

It is contraindicated in patients with known hypersensitivity to Nitazoxanide or any components of the preparation.

 

Side Effects

The most frequent side effects, reported by Nitazoxanide are abdominal pain, vomiting and headache. These side effects are typically mild and transient in nature. Very rare side effects include- nausea, anorexia, flatulence, increased appetite, enlarged salivary glands, increased creatinine & SGPT level, pruritus, rhinitis, sweating, dizziness, discolored urine etc.

 

Pregnancy & Lactation

Nitazoxanide is classified as a pregnancy category B by the US Food and Drug Administration. However, no suitable and well-controlled studies in pregnant women have been conducted. Because animal reproduction studies do not always predict human response, this medication should only be used during pregnancy if absolutely necessary. Human milk has been found to contain nitazoxanide. As a result, Nitazoxanide should be used with caution during lactation.

 

Precautions & Warnings

Patients with hepatic and biliary disease, as well as those with renal disease, should use nitazoxanide with caution.
