Description

Dasacent is a medication primarily used for treating Philadelphia chromosome-positive (Ph+) chronic myeloid leukemia (CML) and acute lymphoblastic leukemia (ALL) in both adult and pediatric patients. 

  • Adults:
    • Newly diagnosed Ph+ CML in chronic phase.
    • Chronic, accelerated, or blast phase Ph+ CML with resistance or intolerance to prior therapy.
    • Ph+ ALL with resistance or intolerance to prior therapy.
  • Pediatric patients (1 year and older):
    • Ph+ CML in chronic phase.
    • Newly diagnosed Ph+ ALL in combination with chemotherapy.
  • Mechanism of Action: Dasatinib inhibits several kinases including BCR-ABL, SRC family, c-KIT, EPHA2, and PDGFRβ. This inhibition helps overcome Imatinib resistance and targets various signaling pathways involved in leukemia progression.
  • Absorption: Peak plasma concentrations are reached between 0.5 to 6 hours after oral administration.
  • Metabolism: Primarily metabolized by CYP3A4, with minor contributions from other enzymes like FMO-3 and UGT.
  • Excretion: Mostly excreted in the feces, with only 4% excreted in urine.
